This course will present an overview of what simulation can bring for the development, optimization and troubleshooting of chromatographic processes. It will focus on applications relevant for biotech industries (e.g., buffers, amino acids, proteins, oligonucleotides).

The course will be balanced between theoretical lectures and hands-on sessions. Theoretical lectures will provide the fundamentals of chromatography modeling, which derives from the understanding and description of the physical phenomena at stake, namely thermodynamics, kinetics of mass transfer and hydrodynamics. In the hands-on sessions, the attendees will have access to the software Ionic®. They will be able to play with the software and perform simulations on their own, in particular to predict the impact of key operating parameters (e.g., crude composition, loading volume, flowrate, gradient slope,…) on process performances.
The training will be presented through practical case studies covering various chromatography techniques: size exclusion, affinity, hydrophobic interaction and ion exchange chromatography. For each type of chromatography, a proper mechanistic model will be presented and the key experiments necessary to identify model parameters will be discussed.
